摘要
目的研究黄芩苷在玻碳电极上的伏安行为,建立牛黄解毒片中黄芩苷的含量测定方法。方法Waters Plus—C18(1mL/50mg,30μm)固相萃取柱进行样品前处理,采用循环伏安法与差示脉冲伏安法研究黄芩苷在玻碳电极上的伏安行为。结果用20%甲醇1.0mL能完全洗脱黄芩苷;在pH3.0磷酸盐缓冲溶液中,黄芩苷的差示脉冲伏安峰电流与其浓度在1×10^-6~9×10^-5mol/L呈良好的线性关系,检测限为1×10^-7mol/L。结论本法简便、经济,灵敏度高,用于牛黄解毒片中黄芩苷的含量测定效果满意。
Objective To study the voltammetric behaviors of baicalin at the glassy carbon electrode and develop a method for the determination of baicalin in Niuhuang Jiedu Plan. Methods The pretreatment of sample was performed with a solid phase extract column, Waters Plus-C18(1 mL/50 mg, 30 μm). The electrochemical behavior of baicalin was investigated by using cyclic voltammetry and differential pulse voltammetry at the glassy carbon electrode. Results Using 25% methanol 1.0 mL as elution solution, the baicalin can be eluted completely from the solid phase extract column. In pH 3.0 phosphate buffer solution, the peak current of baicalin differential pulse voltammetry was dependent linearly on its concentration of baicalin over the range of 1×10^-6 - 9 ×10^-s mol/L with a detection limit of 1 ×10^-7 mol/L. Conclusion It is a convenient, economical and sensitive property for the determination of baicalin in Niuhuang jiedu plan with satisfactory results.
