摘要
目的建立小鼠四氯化碳性及酒精性肝纤维化模型并初步探讨干细胞动员情况,确定骨髓源性肝干细胞的表面标记。方法四氯化碳皮下注射和酒精灌胃法使小鼠形成肝纤维化,分别取其骨髓干细胞,采用流式细胞仪检测肝纤维化模型中不同干细胞标记的细胞群体包括c-kit+Lin-,Thy1.2+Lin-,CD34+Lin-,Sca+1Lin-的数量变化,寻找可能的肝干细胞动员情况。结果与对照组相比,两组纤维化模型小鼠骨髓内c-kit+Lin-细胞显著增高,其他干细胞类型细胞数量变化小。结论c-kit+Lin-细胞的数量在肝纤维化模型小鼠中存在明显动员情况,可能参与肝脏的修复,并成为肝干细胞的标记物。
Objective To establish hepatic fibrosis model induced by alcohol and tetrachloride (CCL4 ) and to investigate the mobilization of bone marrow derived liver stem ceils and the identification of its surface marker. Methods Hepatic fibrosis model was induced by gavage with white spirits and subcutaneou injection of CCL4. Bone marrow cells were subsequently collected. Stem cell surface markers, including c - kit^+ Lin - , Thy1. 2^++ Lin - , CD34^+ Lin - and Sea^+1 Lin - markers were detected by using flow cytometry . Results Compare with the control group, the c - kit ^+ Lin - cells elevated significantly in each of the hepatic fibrosis model models. The other stem cell populations showed little quan- titative changes. Conclusion The c - kit ^+ Lin - ceils are mobilized in hepatic fibrosis and may participated in the repair of liver. The c - kit^+ Lin - cell can be used as marker of liver stem cells.
