摘要
目的:探讨大葱油对胃癌细胞的影响以及诱导胃癌细胞凋亡的机制.方法:MTT法评价大葱油对胃癌细胞增殖的影响.荧光染色观察细胞形态学变化.流式细胞术和放免法检测细胞凋亡及细胞内游离Ca2+和环磷酸腺苷(cAMP)浓度.结果:随着大葱油浓度的增加,作用24h后,细胞增殖抑制率分别达到5.71%,36.33%,44.34%,54.52%和74.15%,IC50≈65μg/mL.荧光显微镜下观察结果显示,实验组出现细胞凋亡形态学变化.100μg/mL大葱油作用于胃癌细胞1~12h,细胞总凋亡率分别为50.16%,53.48%,77.84%,83.71%和41.34%;早期细胞凋亡率分别为50.15%,33.25%,11.69%,4.65%和0.84%.100μg/mL大葱油作用于胃癌细胞5min^12h,各相应时间点细胞内游离Ca2+含量依次为(71.00±6.08),(77.67±1.53),(79.67±1.53),(83.33±5.03),(76.00±6.06),(75.67±2.52),(72.33±4.24)和(65.00±2.00)nmol/L;细胞内cAMP浓度依次为(5.76±0.36),(18.51±2.06),(20.40±1.65),(16.09±1.46),(12.16±0.78),(9.88±0.53),(9.47±0.78)和(9.82±1.06)pmol/mg(P<0.05).结论:大葱油可抑制胃癌细胞增殖,并通过上调细胞内Ca2+和cAMP水平诱导其凋亡.
AIM: To investigate the effects of Allium fistulosum L. oil (ALFO) on gastric cancer cell line and the possible signal molecular mechanism underlying apoptotic process induced by AFLO. METHODS: Human gastric cancer MGC80-3 cells were cultured in vitro and treated by AFLO. Then its growth inhibitory rate was counted and IC50 was measured. Apoptotic process was investigated with fluorescence microscope. Intracellular-free calcium content at different times was determined by flow cytometry. Intracellular cyclic adenosine monophosphate (cAMP) levels were determined by radioimmunoassay (RIA). RESULTS: With increasing the concentration of AFLO, cell growth inhibitory rates went up correspondingly 24 h after treatment with AFLO and reached 5.71% , 36.33% , 44.34% , 54.52% and 74.15% respectively. ICs0 ~65 μg/mL. Hoechst 33258 fluorescent staining showed that apoptosis appeared in some cells treated by AFLO (100 μg/mL). When MGC80-3 cells were incubated in the presence of AFLO ( 100 μg/mL) from 1 to 12 h, the total apoptotic rates were 50. 16%, 53. 48%, 77. 84%, 83. 71% and 41.34%, respectively, and the early apoptotic rates were 50.15% , 33.25% , 11.69%, 4.65% and 0.84%, respectively. In MGC80-3 cells treated by AFLO ( 100 μg/mL) for 5 min to 12 h, free calcium contents were (71.00 ±6.08) , (77.67 ± 1.53), (79.67 ±1.53), (83.33 ±5.03), (76.00 ±6.06), (75.67 ± 2.52 ), ( 72.33 ± 4.24) and (65.00 ± 2.00) nmol/L, respectively and cAMP concentrations were (5. 76 ± 0. 36), (18.51 ±2.06), (20.40 ± 1.65), ( 16.09 ± 1.46), (12.16 ± 0.78),(9.88±0.53),(9.47±0.78),(9.82 ±1.06) pmol/ mg, respectively. CONCLUSION: AFLO can inhibit the growth of gastric cancer cell line MGC80-3 and induce the apoptosis through increasing intracellular Ca^2+ and cAMP levels.
