摘要
目的评价鞘内注射医用臭氧对兔神经行为学和脑、脊髓超微结构的影响,以探讨医用臭氧的神经毒性。方法新西兰大白兔30只,随机分为5组(n=6),穿刺对照组(s组)仅行小脑延髓池穿刺;纯氧对照组(02组)行小脑延髓池穿刺后注射医用纯氧2ml;不同浓度医用臭氧组(O2-O3 30组、O2-O3 50组、O2-O3 80组)行小脑延髓池穿刺后分别注射30、50、80mg/L医用臭氧2ml。分别于麻醉前和注射后24h测定双前足热刺激缩足潜伏期(PWHL)及机械刺激缩足反应阈值(PWMT),并进行运动功能(MF)评分及后肢趾外展(TA)评分。测定神经行为学的各项指标后处死兔,取大脑枕叶皮质和颈脊髓(C1~4)组织,透射电镜下观察超微结构。结果麻醉前及注射后24h时PWHL、PWMT、MF评分及TA评分组间及组内比较差异无统计学意义(P〉0.05)。不同浓度医用臭氧组大脑枕叶皮质及颈脊髓组织均有不同程度的病理损伤。结论鞘内注射医用臭氧30、50和80mg/L(2m1)对兔具有一定程度的神经毒性,且与浓度有关。
Objective To investigate the neurotoxicity of intrathecal (IT) administration of different concentrations of ozone ( 03 ). Methods Thirty New Zealand white rabbits of both sex aged 3-5 months weighing 2.5-3.0 kg were randomly divided into 5 groups ( n = 6 each) : group Ⅰ control; group Ⅱ pure oxygen ( 100% O2); group Ⅲ 70% O2-30% O3; group Ⅳ 50% O2-50% O3 and groupV20% O2-80% O3. The animals were anesthetized with intraperitoneal pentobarbital. Cerebello-medullary cistern was punctured for IT administration of O2 and O3 . Ozone was manufactured by the ozone generator. The front paw withdrawal latency to heat (PWHL) and to mechanical stimulation (yon Frey filament) (PWML), motor function (MF), hind paw abduction (TA) were measured before and one day after IT O3 administration. The animals were anesthetized again after last measurement. Tissues were obtained from occipital lobe and cervical spinal cord for electron microscopic examination. Results IT administration of the 3 concentrations of ozone did not affect PWHL, PWML, MF and TA. Electron microscopic examination showed that occipital lobe and cervical spinal cord were damaged in a concentration dependent manner in group Ⅲ , Ⅳ and Ⅴ . Conclusion Ozone 30, 50 and 80 mg/L (2 ml) administered intrathecally has neurotoxic effects, depending on concentration.
