正常人外周血中CD8^+CD25^+T细胞亚群的数量及其细胞因子表达的初步研究 被引量:4

摘要 目的:研究正常人外周血CD8+CD25+T细胞亚群的数量及其表达IFN-γ、IL-10和TGF-β的细胞数量情况,以期得到一组该亚群细胞正常数值范围,为研究CD8+CD25+T细胞亚群在自身免疫性疾病、过敏性疾病和器官移植中的变化及其临床意义提供参考。方法:分离正常人外周血单个核细胞,利用三色流式细胞术分别检测CD3+T细胞中CD8+CD25+、CD8+CD25-、CD8-CD25+和CD8-CD25-亚群的比例,并计算其细胞数量;并对经过或未经过离子霉素(ionomycin)、佛波醇乙酯(PMA)联合刺激5h的CD8+CD25+T细胞检测表达IFN-γ、IL-10及TGF-β的细胞数量情况。结果:正常人外周血CD3+T细胞中CD8+CD25+细胞亚群约为2.52%,数量达37.09×106/L,经离子霉素、佛波醇乙酯联合刺激培养5h后其比例、数量均略有降低;CD8+CD25+T细胞表面TGF-β阳性率达到93.8%,刺激对其表达无明显影响;但刺激后IFN-γ+T细胞明显增多,而IL-10+的T细胞未见明显升高。结论:正常人外周血CD3+T细胞中存在少量的CD8+CD25+细胞亚群,该细胞表面稳定地高表达TGF-β,可对刺激呈现IFN-γ反应,但不对刺激产生增殖反应。 Objective:In order to know the clinical significance of the CD8^+CD25^+T cell in autoimmune disease,hypersensitivity disease and organ transplantation, the normal limits of this T cell subset was obtained by studying the number ofCD8^+CD25^+T cell and its productions of IFN-γ,IL-10 and TGF-β in the normal peripheral blood. Methods:Mononuclear cells were isolated from healthy donors. The percentages and absolute numbers of CD8^+CD25^+,CD8^+CD25^-,CD8^-CD25^+ and CD8^-CD25^- from CD^3+ T cells were detected by flow cytometry. In addition, stimulating with/without ionomycin and PMA 5 hours later,the expressions of IFN-γ, IL- 10 and TGF-β in the CD8^+CD25^+ T cells was measured. Results:The percentage of CD8^+CD25^+ subgroup in CD^3+ T cells from normal human peripheral blood was 2.52% ,the absolute number was 37.09×10^6/L. The percentage and absolute number decreased after stimulated with ionomycin and PMA 5 hours. The expression of TGF-β on the surface of CD8^+CD25^+ T cells up to 93.8% ,stimulation seems not obviously affected its expression but IFN-γ T cells increased obviously,however,IL-10^+ T cells didn't obviously up-regulal. Conclusion:There were a few CD8^+CD25^+ T cells in CD3^+ T cells from normal peripheral blood. They expressed highly TGF-β in the cell surface,and could have IFN-γ reaction but no proliferation after stimulation.
