摘要
目的采用HPLC-ELSD测定五苓散中泽泻醇A24-乙酸酯、泽泻醇B23-乙酸酯的含量。方法色谱柱:大连依利特Hy-persilC18柱(4.6mm×250mm,5μm);流动相:乙腈-水(75:25);检测器:Alhech2000型蒸发光散射检测器;流速为0.8mL·min^-1;柱温:室温;ELSD气体:高纯氮气;流速为2、0L·min^-1;漂移管温度:82℃。结果在五苓散中测定出的泽泻醇A24-乙酸酯、泽泻醇B23-乙酸酯分别在0.330~1.980μg(r=0.9995),0.624~4.680μg(r=0.9994)内呈良好的线性关系。平均回收率分别为101.4%,99.6%,RSD分别为1.7%,3.0%(n=6)。结论采用本法测定五苓散中泽泻醇A24-乙酸酯、泽泻醇B23-乙酸酯的含量,方法简便、可靠、重复性好。
OBJECTIVE To establish a method for the determination of alisol A24-acetate and alisol B23-acetate in Wulingsan. METHODS A HPLC combined with evaporation light scattering detector(ELSD)was applied. Chromatographic column was Hypersil C18 (4.6 mm×250 ram,5 μm). The mobile phase consisted of acetonitrile-water(75:25 )at the flow rate of 0.8 mL·min^-1. The gas flow rate of ELSD was 2. 0 L · min^-1 and the temperature of drift tube was 82 ℃. RESULTS The linear ranges of the two compo- nents were 0. 330 - 1. 980 μg( r = 0. 999 5, n = 5 ) and 0. 624 - 4. 680 μg( r = 0. 999 4, n = 5 ) respectively. The average recoveries of the two components in Wulingsan were 101.4% ( RSD = 1.7 % ) and 99. 6% ( RSD = 3.0% ) ( n = 6 ) , respectively. CONCLUSION The method is simple ,practical and reliable to be used for the quality control of the Wulingsan.
