摘要
目的:探讨高压氧(HBO)对荷S-180肉瘤小鼠在化学治疗中的增效作用及其机理。方法:240只荷S-180肉瘤的ICR小鼠被随机等分为不治疗对照组、高压氮氧组、HBO组、SOD-HBO组、HBO-平阳霉素组、HBO-环磷酰胺组、平阳霉素组和环磷酰胺组共8个组。分别使用化学治疗结合HBO疗法,采用电子自旋共振(ESR)技术直接检测S-180肉瘤组织中的氧自由基(OFR)含量,并应用分光光度计法测定其脂质过氧化物丙二醛(MDA)的含量,同时对各组动物肉瘤的体积、重量、坏死率及小鼠致死率进行了观察。结果:HBO组、HBO-平阳霉素组及HBO一环磷酰胺组的动物与相应的对照组比,肉瘤组织中的OFR和MDA含量均显著增高(P<005~0.001);同时瘤组织坏死率较高和坏死所需时间较短,且动物存活率较高,肉瘤体积和重量也显著小于对照组(P<0.05~0.001)。结论:荷S-180肉瘤的小鼠经HBO暴露可减缓荷瘤的生长速度并加速肉瘤细胞的坏死,平阳霉素或环磷酰胶结合HBO治疗荷S-180肉瘤的小鼠,能显著增强药物的抗肿瘤作用,同时减轻药物的副作用和延长其生命。
Aim: To clarify the mechanism of the synergetic effect of hyperbaric oxygen (HBO) and chemo-therapy on S-180 sarcoma mice. Methods: 240 ICR mice with S-180 sarcoma were randomly divided into 8 groups, namely the control group, normoxic hyperbaric nitrox(N2-O2) group, HBO group,SOD-HBO group, HBO - pingyangmycin (HBO- P) group, HBO-cyclophosphamidum (HBO-C ) group,pingyangmycin(P) group and cyclophosphamidum (C ) group. The mice with S-180 sarcoma were treated with combined HBO and chemo-therapy. The oxygen free radical (OFR) content in S-180 sarcoma tissue was detected by eletron spin resonance (ESR) technique and the malondialdehyde (MDA) measured by spectrophotometry. The volume, weight and necrosis rate of sarcoma tissue and the fatality of mice in different groups were observed. Results: The contents of OFR and MDA in sarcoma tissue in the HBO group, HBO-P group and HBO-C group were markedly higher than those in the respective control groups (P<0. 05 ~ 0. 001) ; the necrosis rate of sarcoma tissue was higher, the necrosis time needed was shorter, the survival rate of mice was also higher, and the volume and weight of sarcoma tissue were both obviously smaller or lighter than those of the controls(P<0. 05~0. 001) . Conclusion:The results suggested that the mice exposed to HBo may slow the growth and hasten the necrosis rate of S-180 sarcoma tissue. The anti-tumor effect of pingyangmycin or cyclophosphamidum on S-180 sarcoma mice could be enhanced significantly by the combined use of HBO;at the same time the side effects of these drugs lessened, and the lives of mice prolonged.
