摘要
目的建立甲磺酸帕珠沙星的血药浓度的高效液相色谱方法。方法以特拉唑嗪为内标,以ZORBAX C18(4.6mm×150mm,4.6μm)色谱柱为分析柱;采用乙腈-0.01mol.L-1柠檬酸水溶液-三乙胺-冰醋酸(13.9∶85.7∶0.3∶0.1,v/v/v/v,pH值5.5)为流动相;流速为1ml/min;激发波长为335nm;发射波长为450nm。结果甲磺酸帕珠沙星线性范围0.037 2-19.5333μg.ml-1,定量下限为0.0372μg.m^l-1。日内、日间精密度RSD〈8%,平均回收率〉50%。结论本方法简便、准确、灵敏,特异性强,重复性好,适用于甲磺酸帕珠沙星临床药动学研究。
Aim To establish an HPLC method for determination of Pazufloxacinmesilate in human blood serum. Methods Terazosin was used as the internal standard with the ZORBAX C18 (4.6mm × 150mm ,4.6μm) as the analytical column. The mobile phase consisted of acetonitrile-0.01 mol·L^- 1 citric acid-triethylamine-acetum ( 13.9: 85. 7: 0.3: 0.1, v/v/v/v, pH:5.5). The flow rate was 1.0ml/min with fluorescence detection set at λex 335nm and λem 450nm. Results The linear range of the calibration curve was within drug sersum concentrations of 0.037 2 19.533 3μg·ml^-1. The detection limit was 0.037 2μg·ml^-1 ( n =5). The average recovery was above 50%, The inter and intra-day precision ( RSD )was less than 8%. Conclusions The method is shown to be sensitive, accurate, simple and reproducible for the determination of Pazufloxacinmesilate levels in human blood serum. It is suitable for pharmacokinetic study of Pazufloxacinmesilate.
