摘要
目的:建立一种操作简便、可行的 HPLC 法同时测定心可舒制剂中葛根素和丹酚酸 B 的含量。方法:色谱柱:SHIMAD-ZU Shim-pack VP-ODS(250 mm×4.6 mm,5 μm);流动相:乙腈-0.1%磷酸,梯度洗脱:0~15 min 时乙腈-0.1%磷酸比例为13:87,15~45 min 时比例为24:76;流速:1 mL·min^(-1);检测波长:305 nm。结果:葛根素进样量在0.0252~0.5040 μg范围内与峰面积线性关系良好(r=0.9999);丹酚酸 B 进样量在0.0154~0.308μg范围内与峰面积线性关系良好(r=0.9999)。心可舒胶囊中葛根素和丹酚酸 B 平均回收率分别为99.34%和99.57%,心可舒片中葛根素和丹酚酸 B 平均回收率分别为99.08%和100.0%。结论:采用 HPLC 法测定心可舒制剂中葛根素和丹酚酸 B 的含量,样品处理方法简便,测定结果准确,方法专属性强,精密度高,重复性好。
Objective:To propose a simple, convenient, feasible HPLC quantitative method for simultaneous determination of puerarin and lithospermic B in Xinkeshu preparations. Method:Chromatographic column:SHIMADZU Shim - pack VP - ODS column (250 mm × 4.6 mm ,5 μm particle size) ;Mobile phase : acetonitrile and 0. 1% phosphoric acid ,gradient elution :0 -15 min acetonitrile and 0. 1% phosphoric acid( 13:87 ) and 15 -45 min acetonitrile and 0. 1% phosphoric acid (24:76 ) ; Flow rate : 1 mL · min^ - 1 ; Wavelength : 305 nm. Results: The linear range of puerarin was good within the range of 0. 0252 -0. 504 μg( r = 0. 9999 ) and that of lithospermic B was good within the range of 0. 0154 - 0. 308 μg ( r = 0. 9999 ). The average recoveries of puerarin and lithospermic B were 99. 34% and 99. 57% in Xinkeshu capsules, and the average recoveries of puerarin and lithospermic B were 99.08% and 100. 0% in Xinkeshu tablets. Conclusion:The method is simple, accurate, stable and reliable for the simultaneously determination of puerarin and lithospermic B in Xinkeshu preparation.
