黄芪合丹参注射液对急性脑出血大鼠神经细胞凋亡表达的影响 被引量:19

摘要 目的:观察黄芪合丹参注射液对急性脑出血大鼠神经细胞凋亡表达的影响。方法:104只SD雄性大鼠随机分为正常组、假手术组、模型组、丹参组、黄芪合丹参组。黄芪合丹参组在制成脑出血模型后腹腔注射黄芪注射液(3.4g.kg-1.d-1)和丹参粉针溶解液(69mg.kg-1.d-1)。测试大鼠的行为学,对脑组织行末端脱氧核糖核酸转移酶介导的缺口末端标记法(TUNEL法)染色及Bcl-2蛋白免疫组化染色,观察并计数血肿周围阳性细胞表达。结果:术后3、7天,黄芪合丹参组神经功能缺损评分低于丹参组及模型组(P<0.01)。术后1、3、7天,黄芪合丹参组TUNEL阳性细胞数低于模型组和丹参组(P<0.01或P<0.05),Bcl-2蛋白阳性细胞数高于模型组(P<0.01或P<0.05)。结论:黄芪合丹参能抑制脑出血大鼠神经细胞的凋亡,其中通过促进Bcl-2蛋白的表达是抑制细胞凋亡的机制之一。 Objective: To observe the effect of Astragalus mongholicus and Salvia miltiorrhiza injection on expression of nerve cells apoptosis in acute intracerebral hemorrhage rats.Methods: 104 Sprague-Dawley male rats were divided into normal group,sham operation group,model group,Salviae miltiorrhiza group,Astragalus mongholicus and Salviae miltiorrhiza group at random.In group of Astragalus mongholicus and Salviae miltiorrhiza,Astragalus injection(3.4g·kg^-1·d^-1) and Salviae miltiorrhiza injection(69mg·kg^-1·d^-1) was injected after the intracerebral hemorrhage model was established.Rats' learning and memory were tested.Terminal deoxynucleotidyl transferase mediated nick end labeling(TUNEL),was used to detect the never cells apoptosis.The protein expression of Bcl-2 was detected with immunohistochemistry staining.Results: On the third,seventh day after operation,the score of neural functional defect of Astragalus mongholicus and Salviae miltiorrhiza group was lower than that of Salvia miltiorrhiza group and normal group(P〈0.01).On the first,third,seventh day after operation,the number of TUNEL-positive cells around hematoma of Astragalus mongholicus and Salviae miltiorrhiza group was fewer than that of model group and Salviae miltiorrhiza group(P〈0.01 or P〈0.05).And compared with model group,the number of Bcl-2 protein positive cells of Astragalus and Salviae miltiorrhiza group increased significantly(P〈0.01 or P〈0.05).Conclusion: Astragalus mongholicus and Salviae miltiorrhiza injection can inhibit neural cells apoptosis in intracerebral hemorrhage rats by promoting expression of Bcl-2 protein.
