摘要
目的探讨地氟醚对内毒素(LPS)诱导大鼠肺组织氧化应激反应的影响。方法雄性Wistar大鼠48只,体重200~290 g,随机分为4组(n=12),对照组(C组)股静脉注射生理盐水1.2ml后机械通气4 h;LPS组(L组)股静脉注射LPS 5mg/kg后机械通气4 h;地氟醚1.0MAC组(D1)和地氟醚1.5 MAC组(D2)组:股静脉注射LPS 5 mg/kg后机械通气,分别吸入地氟醚1.0 MAC和1.5 MAC 4 h,机械通气4 h后处死大鼠,测定肺组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、一氧化氮(NO)含量、总一氧化氮合酶(tNOS)活性、诱导型一氧化氮合酶(iNOS)活性、iNOS mRNA和iNOS表达水平。结果与C组比较,L组SOD活性下降,MDA含量、NO含量、tNOS活性、iNOS活性、iNOS mRNA和iNOS表达升高(P〈0.01);与L组比较,D1组上述各项指标差异无统计学意义(P〉0.05),D2组SOD活性下降,MDA含量、iNOS活性、iNOS mRNA和iNOS表达升高(P〈0.05);与D1组比较,D2组SOD活性下降,MDA含量、iNOS活性、iNOS mRNA和iNOS表达升高(P〈0.05)。结论吸入地氟醚1.5 MAC 4 h可加重LPS诱导大鼠肺组织氧化应激反应,吸入地氟醚1.0 MAC对其无影响。
Objective To investigate the effects of desflurane on pulmonary oxidative stress response induced by endotoxin in rats. Methods Forty-eight male Wistar rats weighing 200-290 g were randomly divided into4 groups (n = 12 each): Ⅰ control group (C); Ⅱ LPS group received LPS 5 mg/kg intravenously; Ⅲ ,Ⅳ desflurane group (D1 and D2 ) received LPS 5 mg/kg followed by 4 h 1.0 and 1.5 MAC desflurane inhalation respectively. All animals were anesthetized with intraperitoneal pentobarbital 100 mg/kg, tracheostomized and mechanically ventilated for 4 h after IV LPS or normal saline injection. Right carotid artery and left femoral vein were cannulated for BP monitoring and fluid and drug administration. The animals were killed at the end of 4 h mechanical ventilation by exsangnination. The lungs were immediately removed for determination of MDA and NO content, SOD and total nitric oxide synthase (tNOS) and inducible NOS activities and iNOS mRNA and protein expression in the lung tissue. Results The SOD activity was significantly lower and the MDA and NO content, tNOS, iNOS activity and iNOS mRNA and protein expression were significantly higher in the lung tissue in LPS group than in control group. There were no significant differences in these variables between group LPS and D1, The SOD activity was significantly lower, the MDA content the iNOS activity and iNOS mRNA and protein were significantly higher in the lung tissue in group D2 than in group LPS and D1 . Conclusion Four hour 1.5 MAC desflurane inhalation is detrimental to the endotoxin-induced pulmonary oxidative stress response in rats while 4 h 1.0 MAC desflurane inhalation is not.
