叶下珠有效部位总提取物对比格犬的长期毒性实验

摘要 目的观察叶下珠有效部位总提取物长期毒性实验可能造成对机体的毒性反应。方法叶下珠有效部位总提取物470,210,90 mg/kg每天口服给药1次;连续给药45,90,180 d及停药后30 d,观察对比格(Beagle)犬生长发育以及血液学常规、血液生化指标、心电图的影响;以及连续给药180 d和停药后30 d对主要脏器体重比、主要脏器病理形态的影响。结果对生长发育、血液学常规、心电图均无影响。90 mg/kg:连续给药90 d,BUN,TP,Glu升髙明显,T-CHO下降;但给药180 d及停药后30 d均恢复正常。210,470 mg/kg:ALT,ALP,Glu均显著升髙,TP,Alb,T-CHO显者下降;肝细胞轻度浊肿、空泡变性。但上述毒性改变均于停药后30 d恢复正常。结论90 mg/kg组为安全剂量;210 mg/kg以上剂量为中毒剂量;停药后可恢复正常。 Objective To observe the chronic toxicity of TEY in Beagle dog. Methods Rats of three experimental groups were i. g. TEY at dose of 1280 mg/(kg · d) ,640 mg/(kg · d)and 320 mg/(kg· d), respectively,6 days per week for 6 months. Rats of control group were given distilled water at the same volume. Growth of rats were observed. Indexes of hematology and blood biochemistry were measured. Ratio of viscera to body were measured and the major organs were examined by the pathology. All these tests were carried out three and six months after i. g. and one month after stopping i. g. Results The outer appearance and behavior, weights, the indexes of hematology and blood biochemistry were normal. The organ coefficient, the cardiogram, the pathological examination had no significant difference by comparing with control group. Conclusion TEY has no significant toxicity for long - term administration to rats.
