摘要 目的探讨苦参碱对A549细胞增殖和PinX1mRNA表达的影响。方法用不同浓度的苦参碱作用A549细胞48h,流式细胞仪检测增殖指数,RT-PCR检测PinX1 mRNA表达。结果随着苦参碱浓度的增加(0mg/ml、0.5mg/ml,1.0mg/ml、1.5mg/ml、2.0mg/ml),A549细胞增殖指数(PI)逐渐下降(34.35±1.26、18.34±1.01、13.38±1.02、8.47±0.86、7.01±0.75),PinX1mRNA表达逐渐增强(PinX1/β-actin相对灰度值0.42、0.81、1.44、2.05、2.53),二者呈明显负相关(r=-0.89,P<0.05)。结论苦参碱呈浓度依赖性抑制A549细胞的增殖可能与增强PinX1mRNA的表达有关。 Objective To observe the effects of matrine on proliferation and PinX1 rnRNA expression in lung adenocarcinoma cell line A549. Methods After matrine (0,0. 5,1.0,1.5,2. 0mg/ml) was used to treat A549 cells for 48h in vitro, the proliferation index (PI) was detected by flow cytometry and the expression of PinX1 mRNA was detected by RT-PCR Results With the increasing concentration of matrine, PI decreased by (34. 35 ± 1.26), (18. 34 ± 1.01), (13.38 ± 1.02), (8. 47 ± 0. 86), (7. 01 ± 0. 75), and the expression of PinX1 mRNA increased gradually (PinX1/β-actin: 0. 42,0. 81,1.44,2. 05,2. 53). PI had significant negative correlation with the expression of PinX1 rnRNA in A549 cells (r= - 0. 89,P〈0. 05). Conclusion Matrine inhibited the proliferation of A549 cells in vitro probably by enhancing the expression of PinX1 mRNA.
