摘要
目的:为研究在靶肌肉上注射神经生长因子(Nervegrowthfactor,NGF)的可行性。方法:用硅胶管桥接大鼠双侧5mm长坐骨神经缺损,术后第1天向右侧胫前肌及腓肠肌分别注射高剂量(8μg/d)、低剂量(0.8μg/d)的2.5sNGF和等量生理盐水,术后20、40、80天,运用胫前肌湿重、运动神经传导速度测定、形态学及图像分析等手段测定各项指标。结果:高NGF组和低NGF组再生神经在结构及功能上均优于对照组,术后40天高NGF轴突直径优于低NGF组,高NGF组内右侧明显优于左侧。结论:靶肌肉注射NGF能促进鼠坐骨神经再生。
Objective:The effect of targeted muscle injecting nerve growth factor(NGF) was examined in the neural repairment of adult rat sciatic nerves.Methods:Forty five SD rats were divided into three groups:high dosage NGF group, low dosage NGF group and control group.The sciatic nerves of rats were severed forming a 5 mm gap and were bridged by sclicon tubing.M.tibialis anterior and M.gastrocnemius were used as injecting positions and high dosage NGF group was given 8μg 2 5s NGF,low dosage NGF group was given 0 8μg 2.5s NGF,and control group was given normal saline every day after operation.The animals were sacrificed at intervals of 20 days,40 days and 80 days respectively.Morphology and fuctional recovery of nerve trunk regeneration was observed by means of weighting the wet weight of M.tibialis anterior,measuring the motor nerve conduction velocities (MNCV),conducting microsocopic examination and axon video analysis.Results:Both experimental groups excelled control group and among high dosage NGF group the right side excelled left side in the morphology and fuctional of regenerating nerve,high dosage NGF group excelled low dosage NGF group in diameter of regenerating axon at 40th day postoperative.Conclusion:It was first demonstrated that targeted muscle injecting NGF can enhance rat sciatic nerve regeneration.
