摘要 以孝感种源的3 a生香椿苗木为材料,分析了基本培养基对茎段组织培养的影响,研究了香椿茎段组织培养以及无菌苗的移栽技术。结果表明:0.1%HgCl2对香椿茎段的消毒效果较好;MS比1/2 MS、B5培养基更适于茎段的萌芽培养,萌芽率达80%。MS+1.0 mg/L BA+0.5 mg/L GA3+0.5 mg/L KT为最佳增殖培养基,增殖系数达到4.7。生根试验以1/2 MS+1.5 mg/LIBA+30 g蔗糖作为生根培养基较为适宜,生根率达到90%;在以河沙为基质时,移栽成活率达72.5%。 In this study, stem from three years old Chinese toon from Xiaogan was used as material to study the tissue culture and regeneration system in Toona sinensis. The results are as follows: the solution of 0. 1% HgCl2 was efficient for surface sterilizing of the stem;the culture medium with MS was fit for initiate culture, and the germination rate reached 80%. The optimum media for multiplication was MS plus with 1.0 mg/L BA, 1.5 mg/L GA3 and 0. 5 mg/L KT, by which the multiplicated rate was up to 4. 7. Rooting percentage was 90% on 1/2 MS medium added with 1.5 mg/L IBA and 30 g/L sucrose. The plantlets were transplanted into transplanting media with a survival rate of 72, 5%.
