摘要
目的观察鹿瓜多肽注射液对骨折愈合过程中骨形态发生蛋白2(BMP-2)、转化生长因子(TGF-β1)和血管内皮生长因子(VEGF)等生长因子表达的影响,了解鹿瓜多肽注射液促进骨折愈合的分子生物学机制。方法60只新西兰大白兔随机分为三组,制成双侧桡骨中段3mm骨缺损,三组术后每日分别予以肌肉注射0.1ml/kg、0.2ml/kg鹿瓜多肽注射液和0.2ml/kg生理盐水。术后1、2、4、6周分批处死动物,行组织学免疫组织化学观察。结果①不同剂量的鹿瓜多肽注射液均能使骨折处的BMP-2、TGF-β1和VEGF表达量在各时间段较生理盐水组明显增高(P<0.05);②随着使用剂量的增加,BMP-2、TGF-β1和VEGF的表达量也增加,且表达的高峰期提早。结论鹿瓜多肽注射液能调节骨折愈合过程中BMP-2、TGF-β1和VEGF的合成和分泌,进而调节骨原细胞的增殖和成骨细胞、软骨细胞的分化。
Objective To observe the expression of bone morphogenic protein - 2 (BMP - 2), transforming growth factor- β1 (TGF- β1 ) and vascular endothelial growth factor (VEGF) in fracture healing after treated with Cervus and cucumis polypeptide injection so as to find out its action mechanism. Methods Sixty adult New Zealand rabbits were used to produce models with a 3mm defect in the middle of radius and divided into three groups randomly, which were subjected to intramuscular injection of cervus and cucumis polypeptide injection (0.2 ml/kg, 0.1 ml/kg), or physiological saline respectively after operation. Rabbits were sacrificed at the 1 st, 2 nd, 4 th, 6 th week postoperatively and specimens were possessed for histological observation and immunohistochemical observation on staining of BMP, TGF- β1 and VEGF. Results ①The expression of BMP - 2, TGF- β1 and VEGF in cervus and cucumis polypeptide injection groups was higher than the physiological saline group in fracture healing ( P 〈 0,05 ). ② The expression of BMP - 2, TGF - β1 and VEGF in high dose group was higher and earlier than low dose group. Conclusion Cervus and cucumis polypeptide injection could actually promote the production and exudation of BMP - 2, TGF- β1 and VEGF, and modulate proliferation of osteogenetic cells and differentiation of chondrocytes.
