溃疡性结肠炎大鼠淋巴细胞的过渡归巢与CCR7的表达 被引量:7

Hyperactive lymphocyte homing and expression of CCR7 in experimental rats with ulcerative colitis

摘要 目的探讨实验性溃疡性结肠炎(UC)大鼠淋巴细胞过渡归巢现象与淋巴细胞表面CCR7受体表达的关系。方法40只SD大鼠随机分为对照组及模型组,观察结肠黏膜下淋巴细胞过渡归巢现象,逆转录-聚合酶链反应(RT-PCR)半定量法检测结肠组织中CCR7的表达;流式细胞法(FCM)测定结肠回流血液中CCR7+淋巴细胞的比例。结果模型组大鼠结肠黏膜下存在淋巴细胞过渡归巢,派尔集合淋巴结增生,最大截面积比较,差异有统计学意义[(9.56±0.32)比(1.48±0.58),t=5.12,P<0.01],CCR7 mRNA的表达量与对照组比较明显升高[(0.772±0.108)比(0.126±0.029),t=4.37,P<0.01]、模型组结肠静脉回流血中CCR7+淋巴细胞的比例明显高于对照组[(76.58±9.73)%比(16.62±7.84),t=3.97,P<0.01]。结论溃疡性结肠炎大鼠的结肠黏膜下存在淋巴细胞过渡归巢现象,结肠组织中归巢受体CCR7的表达增高,结肠回流血液中CCR7+的淋巴细胞的比例增高。 Objective To investigate the relationship between hyperaetive lymphocyte homing and the expression of CCR7 in experimental rats with ulcerative eolitis. Methods Forty Spague-Dawley rats were divided into eontrol and model groups at random. Hyperactive lymphocyte homing and the CCR7 expression were deteeted by HE and RT-PCR in their eolon tissues respeetively. The CCR7 positive lym- phoeytes from eolonie vein were measured by flow eytometer. Results Hyperaetive lymphocyte homing and hyperplasia of perye' s Patehes were found in model group. The maximum aereage of perye' s Patehes within sliees showed a significant increase in model group as eompared with the eontrol group ( 9.56 ± 0.32 vs 1.48±0.58 ,P 〈0.01 ). The expression of CCR7 mRNA within eolonie tissue was inereased signifieanfly in model group as eompm'ed with the eontrol group, and so were CCR7 positive lymphoeytes (0.772± 0. 108 vs 0. 126 ± 0. 029 and 76.58 ± 9.73 vs 16.62 ±7.84 respeetively,both P 〈 0.01 ). Conduston The phenomena of hyperactive lymphocyte homing has been proved in experimental rats with ulcerative colitis. The up-regulation of CCR7 was also detected in colonic tissue and colonic vein.
