摘要
目的探讨产生两种小鼠恐惧记忆差异的原因。方法选用昆明种(KM)和C57BL/6小鼠,测定血浆皮质酮水平以及检测其差异,并与行为学结果进行相关性比较。结果KM小鼠对照组和训练组血浆皮质酮水平分别为(101.083±70.266)μg/L、(215.453±60.276)μg/L,两者比较差异有显著性(P〈0.01);训练组血浆皮质酮水平与训练结果木僵反应时间(36.00±29.292)s之间无相关性(r=-0.273,P〉0.05)。C57BL/6小鼠对照组和训练组血浆皮质酮水平分别为(108.009±20.452)μg/L、(176.933±38.202)μg/L,两者比较差异有显著性(P〈0.01);训练组血浆皮质酮水平与训练结果木僵反应时间(257.50±36.450)s之间无相关性(r=-0.462,P〉0.05)。两种小鼠对照组、训练组比较,差异无显著性(P〉0.05)。结论KM小鼠与C57BL/6小鼠恐惧记忆差异可能与血浆皮质酮水平无关。
Objective To clarify the roles of hypothalamus-hypohysis-adrenocortical axis played in the interstrain difference in the retention of fear memory in two strains of mice, Kunming mice ( KM ) and C57BL/6. Methods The conditioning was to associate a footplate shock with a tone signal. The duration of tone-induced stupor response 24-hr after the training represents the retention of fear memory. Serum corticosterone (CORT) level was assayed using HPLC method. Results In KM, serum CORT concentration was(215. 453 ± 60.276)μg/L in trained group, significantly higher than that in the control group ( 101. 083±70. 266)μg/L, P〈0.01 ). In C57BL/6, tserum CORT concentration in trained group and the control group were ( 176. 933 ± 38. 202 ) μg/L and (108. 009 ±20. 452)μg/L respectively( P〈 0.01 ). In trained KM, stupor response duration was (36.00 ± 29. 292) s, and in trained C57BL/6, the duration was (257.50 ± 36. 450) s, which were not correlated to the corresponding serum CORT levels( r = - 0. 462, P 〉 0.05 and r = - 0. 273, P 〉 0.05 respectively). Conclusion Fear conditioning induced comparable stress responses in KM and C57BL/6 mice;and serum CORT level might not account for the interstrain difference in retention of fear memory in these two strains.
