摘要
目的观察胚胎脊髓提取液(fetal spinal cord extracts,FE)对神经干细胞(neural stem cells,NSC)分化的影响并探讨其机制。方法制备FE加入NSC培养体系中诱导分化,分别于分化后第3、7天进行免疫荧光细胞化学鉴定,观察分化细胞表型并计算神经元样细胞所占的比例。结果两组均可观察到神经元,实验组所占比例较高,可达23.39%,明显多于对照组(P<0.01)。两组GFAP阳性星形胶质细胞分化已经比较成熟,也可检测出MBP阳性少突胶质细胞。结论FE通过提供胚胎脊髓源性神经营养因子、各种细胞因子和细胞外基质,支持NSC成活并诱导其分化为神经元、星形胶质细胞和少突胶质细胞,且神经元的分化比例达到23.39%。
Objective To evaluate the influence of fetal spinal cord extracts ( FE ) on the differentiation of neural stem cells (NSC) ,and investigate the potential mechanism. Methods The NSC were co-cuhured with FE for 3 or 7 days, the differentiation of these cells was stained by anti-NSE,GFAP and MBP immunocytochemical procedures so as to observe the ratio of neurons. Results FE can induce NSC to differentiate into neurons, astrocytes and oligodendrocyte, the ratio of neurons achieved to 23.39%. Conclusion FE can provide helpful neurotrophic factor, neurochemistry factors or extracellular matrix, create suitable embryonic microenvironment, support the survival of NSC, induce cell differentiation into neurons, astrocytes and oligodendrocyte and the ratio of neurons is 23.39%.
