摘要 目的建立萃取和纯化龙胆中龙胆苦苷的优化参数。方法以龙胆苦苷含量和解吸率为指标,采用微波辅助萃取、大孔树脂吸附纯化和高效液相色谱法测定龙胆苦苷的含量。结果微波辅助萃取优化条件:萃取剂60%乙醇,流速3ml·min-1;大孔树脂分离富集优化条件为:上样速度2.0ml·min-1,洗脱液50%乙醇,流速1.0ml·min-1时分离纯化效果好。结论在优化的萃取、纯化条件下制得这批龙胆原料龙胆苦苷平均含量为34.57mg·g-1。 Objective To investigate the optimum parameters of extraction and purification of gentiopicrin in Gentiana scabra Bunge. Methods Microwave - assisted flow and macroporous absorptive resin were employed respectively in the extraction and purification processes. The content of gentiopicrin in the fractions was evaluated by HPLC. Results The appropriate extraction conditions were : extractant concentration 60% (V/V) ethanol, flow rate 3 ml·min^-1. When the flow rate of sample was 2.0 ml·min^-1, the eluant was 50% (V/V)ethanol and flow rate was 1.0 ml·min^-1 , the effect of separation and purification was satisfactory. Conclusion Under the above optimum conditions the content of gentiopicrin was 34.57mg·g^-1 from this batch of Gentiaria scabra Bunge.
