摘要
目的:探讨雌激素对肠缺血再灌注(Ischemia-Reper-fusion)复合内毒素血症大鼠肠道功能的影响。方法:健康成年雄性SD大鼠72只,按随机化原则分成9组:即假手术组、脓毒症3 h、6 h1、2 h、24 h组(肠系膜上动脉夹闭后再灌注1 h后给内毒素5 mg/kg)和雌激素3 h、6 h1、2 h2、4 h组(造模完毕后立即经肌肉内注射苯甲酸雌二醇注射液500μg/kg)。剖杀各组大鼠,分别测血中肠屏障功能指标二胺氧化酶(DAO)、D-乳酸含量,同时观察小肠病理形态学改变。结果:脓毒症造模后,血浆二胺氧化酶(DAO)、D-乳酸含量均明显升高(P<0.05),小肠显示严重病理损害。给予雌激素后,血浆二胺氧化酶(DAO)、D-乳酸含量显著降低(P<0.05),小肠病理学损伤明显减轻。结论:肌肉内注射雌激素对大鼠肠缺血再灌注复合内毒素所引起的肠屏障功能的损伤有一定的保护作用。
Objective:To investigate the protective effects of estrogen on guts after ischemia-rcperfusion combined with endotoxin challenge in rats. Methods: Seventy-two adult male Sprague-Dawley rats weighing 180 220g were randomly divided into nine groups (sham-operation group, sepsis group of 3 h, 6 h, 12 h, or 24 h and estrogen treatment group of 3 h,6 h, 12 h or 24 h ) with 8 animals in each. A rat model of sepsis was reproduced by ischemia-reperfusion combined with endotoxin challenge, and animals in treatment groups were injected by estrogen with a dose of 500 μg/kg inmlediately after the model of sepsis was made. The rats in each group were killed for determining the levels of diamine-oxidaese(DAO), D-lactic acid(D-LAC) and pathological changes of the intestinal tissues as well. Results: In sepsis groups, the levels of DAO and D-LAC in plasma were increased dramatically, and the intestine exhibited severe pathological lesion. On the contrary, the levels of DAO and D-LAC in plasma were decreased dramatically in estrogen treatment groups with less pathologic signs in the intestine Conclusion : The findings suggest that estrogen may be beneficial to improving intestinal function after sepsis induced by ischemia-repefusion combined with endotoxin challenge in rats.
