成骨生长肽对大鼠成骨细胞骨架蛋白的影响 被引量:2

摘要 目的研究成骨生长肽OGP在体外对大鼠颅盖骨成骨细胞细胞骨架的影响。方法在体外培养的新生大鼠颅盖骨成骨细胞中加入浓度为10-8mol/L的OGP,处理30min,6、12h后收集细胞爬片,采用BODYPY-Phalloidin对骨架蛋白直接荧光染色,在激光扫描共聚焦显微镜下观察OGP作用后,不同时间点成骨细胞细胞骨架的变化情况。结果OGP作用于成骨细胞后,细胞骨架出现解聚重组现象,在6h时解聚作用最为明显,12h后可见细胞骨架发生重组。结论OGP可促进成骨细胞细胞骨架的重组。 Objective To elucidate the effect of osteogenic growth peptide(OGP) on the cytoskeleton in cultured rat osteoblastlike cells. Methods Osteoblast-like cells were treated with 10-8mol/L of OGP for 30min, 6h and 12h respectively. Then cells were processed for BODYPY-Phalloidin direct fluorescence examination of the actin filaments. Confocal laser scanning microscopy was used to investigate the changes of cytoskeleton. Results Disintegration and reorganization of actin filaments were observed in rat osteoblast-like cells treated by OGP at 10^-8mol/L. Disintegration of actin filaments was most obvious after 6h treatment. Actin fil- aments were reorganized after 12h exposure, Conclusion OGP at 10^-8mol/L may stimulate actin cytoskeletal reorganization in rat osteoblast-like cells in vitro.
