摘要
目的观察葡萄糖酸锌(ZnG)、硫酸镁(MgSO4)及两者联合作用对二氧化硅(SiO2)致中国仓鼠肺(CHL)成纤维细胞DNA损伤的的影响,寻找两者的最佳剂量组合。方法常规培养CHL成纤维细胞,按实验分组:①生理盐水对照组(阴性对照);②SiO2组:含SiO2100 mg/L;③ZnG(2.25 mg/L)组;④MgSO4(2.0μmol/L)组;⑤SiO2+ZnG组:内含100 mg/L SiO2外,含ZnG 1.0、1.5、2.25 mg/L;⑥SiO2+MgSO4组:内含100 mg/LSiO2外,含MgSO40.5、1.0、2.0μmol/L;⑥ZnG、MgSO4联合作用组:每组内含SiO2100 mg/L外,采用2因素3水平正交设计,ZnG+MgSO4共设9个剂量组。培养2 h后收获细胞,通过彗星试验检测CHL成纤维细胞DNA损伤情况。结果与SiO2组比,ZbG、MgSO4可以显著减轻SiO2致DNA链断裂作用(P<0.05);1.5 mg/L ZnG与2.0μmol/L MgSO4联合作用效果最好。结论ZnG、MgSO4及两者联合应用在体外可有效减轻SiO2粉尘致CHL成纤维细胞的DNA损伤作用。
[ Objective] To investigate the effect of zinc gluconate and magnesium sulfate in treating DNA damage induced by silica dioxide in Chinese hamster lung cells, [ Methods] Fibroblast cells were cultured with silica and divided into several groups, namely, the SiO2 group, ZnG ( 2.25 mg/L) group, MgSO4 ( 2.0μmol/) group, SiO2 + ZnG group, SiO2 + MgSO4group, ZnG + MgSO4 group and the control; DNA damage was measured with comet assay. [ Results] Compared with the SiO2 group, ZnG + MgSO4 could significantly alleviate DNA damage induced by SiO2( P 〈0.05) ; the combination of 1.5 mg/L ZnG and 2.0μmol/L MgSO4 was proved to be most effective. [ Conclusion] The application of ZnG, MgSO4 and their combination can effectively alleviate DNA damage of CHL cells induced by SiO2 dust.
