摘要
目的探讨复合性刺激导致胃溃疡对大鼠延髓内脏带(MVZ)NSE表达的影响,以及雷贝拉唑预处理后的变化。方法将48只大鼠随机分为应激组(20只)、预处理组(20只)和对照组(8只)。先建立动物模型:应激组和预处理组于应激前半小时分别用生理盐水和雷贝拉唑灌胃,大鼠在(19±1)℃水中力竭性游泳1h后,四肢捆绑在鼠板上垂直浸入冷水中6h,对照组1ml生理盐水灌胃不加应激。延髓冰冻切片后用ABC法检测NSE的表达,胃窦部行石蜡切片HE染色。结果应激组胃黏膜有局灶性出血和脱落,并且MVZ内观察到NSE阳性神经元,预处理组胃黏膜病理改变不显著,MVZ的NSE阳性神经元数量明显减少,对照组胃黏膜和MVZ基本正常。结论①力竭性游泳加冷束缚应激可导致胃黏膜产生应激性溃疡;②应激性溃疡与MVZ的诱导有关联性;③雷贝拉唑能有效预防应激性溃疡的发生,并推测可能参与了MVZ的调节过程。
Objective To explore the expression of NSE in MVZ of rats under stress by compound stimu- lation and the impact of pretreatment by Rabeprazole. Methods The rats of stress group and pretreatment group were respectively garaged NS and Rabeprazole in half an hour before the stress, which was carried out by following steps, the animals were let to exhaustively swim in cold(19±1)℃ water lh and then the estrerni- ties of animals were tied and vertically inundated in the same water for 6 hours.The contrast group was only garaged lml NS The expression of NSE in frozen section of Medullary was detected by immunohistochemistry staining and the pathological changes of antrum were observed by HE staining. Results Compared to the con- trast group, the stress group showed that obvious pathological changes in gastric mucosa and the expression of NSE was significant increased in MVZ. The injury of antrum and MVZ was not remarkable in the pretreat- ment group. Conclusion ①Compound stimulation induced to stress ulcer. ②The induction of MVZ was relevant to stress ulcer. ③Rabeprazole could effectively prevent stress ulcer and it maybe related to the adjustment process of MVZ.
