PMA和PD98059对全反式维甲酸抑制HepG2细胞表达OPN的影响

摘要 目的研究佛波醇酯(PMA)和细胞外信号调节蛋白激酶(ERK1/2)抑制剂PD98059对全反式维甲酸(ATRA)体外抑制人肝癌细胞株(HepG2)表达骨桥蛋白(OPN)的影响及其机制的初步探讨。方法PMA、PD98059分别处理经ATRA处理的肝癌细胞HepG2,光镜下观察细胞形态,West-ernblot检测骨桥蛋白(OPN)、ERK1/2表达及其磷酸化变化,ImagePro软件分析各条带灰度值变化并做统计学分析。结果在ATRA显著抑制肝癌细胞表达OPN并诱导细胞分化的基础上,PMA可刺激OPN表达,这种作用可被PD98059明显抑制。结论激活ERK1/2通路可逆转ATRA抑制HepG2表达OPN的作用。 Objective To investigate the effect of PMA and PD98059 (a specific inhibitor of ERK1/2) on the expression of osteopontin in human hepatoma HepG2 cells treated with all-trans-retinoic acid (ATRA) . Methods HepG2 was treated with/without ATRA, and the group which pretreated by ATRA was then added with PMA and PD98059. The morphology of HepG2 was observed by light microscope and the expression of OPN, P-ERK1/2. ERK1/2 were detected by Western-blots. The gray scale was calculated with Image-Pro. Results HepG2 cells constitutively expressed low level of osteopontin which treated with ATRA for 70days. Application of PD98059 caused a dramatic reduction of PMA induced osteopontin expression. Conclusion Activiation of ERK1/2 pathway can reverse the inhibition of the exprssion of OPN by ATRA in hepG2 cells.
