hTR反义寡核苷酸对食管癌细胞端粒酶活性、细胞凋亡及凋亡诱导因子表达的影响 被引量:2

摘要 目的:探讨封闭端粒酶RNA功能区的反义寡核苷酸(hTR ASODN)对食管癌EC9706细胞端粒酶活性、细胞凋亡及凋亡诱导因子(AIF)表达的影响。方法:应用hTR ASODN1次/d、连续7d转染EC9706细胞,以未加ASODN的脂质体对照组为空白对照,分别于转染后1d、3d、5d、7d采用TRAP-银染法检测EC9706细胞的端粒酶活性,原位末端转移酶标记法及吖啶橙荧光染色法检测EC9706细胞的凋亡及免疫细胞化学法检测AIF蛋白的表达。结果:与空白对照比较,hTR ASODN转染后EC9706细胞端粒酶活性受到抑制,细胞凋亡率及凋亡指数明显升高,AIF蛋白阳性表达率升高(P<0.05)。结论:hTR ASODN可有效抑制EC9706细胞的端粒酶活性,诱导肿瘤细胞凋亡,在此凋亡过程中,AIF可能发挥着重要作用。 Aim : To explore the effects of human telomerase RNA(hTR) antisense oligodeoxynucleotide (ASODN) on the telomerase activity, cell apoptosis, and the expression of apoptosis inducing factor (AIF) of EC9706 cells. Methods: After transfection by hTR ASODN for 1 d, 3 d, 5 d, and 7 d (liposomes without ASODN as control group) , the telomerase activity of EC9706 cells was determined using TRAP, the cell apoptosis was detected using TUNEL and acridine orange fluorescent staining, and the expression of AIF was detected using immunocytochemistry method. Results: After treatment by hTR ASODN, the telomerase activity was inhibited,and the apoptosis rate, the apoptosis index and the positive rate of AIF were increased (P 〈 0.05). Conclusion: hTR ASODN can effectively inhibit the telomerase activity and induce cell apoptosis of EC9706 cells,in which,AIF may play an important role.
