摘要
通过正交实验对松花粉中水溶性多糖水提取条件进行研究,确定松花粉多糖的最佳提取工艺条件:松花粉在料液比为1∶15,温度85℃的热水中浸提2h,浸提次数4次,经大孔树脂脱色,真空浓缩,酶-Sevage结合法除蛋白后,95%乙醇沉淀得花粉粗多糖,苯酚-硫酸法于490nm下测吸光值,计算总糖含量。在以上条件下,松花粉多糖的得率可达1.70%。其中浸提温度、料液比、提取次数、浸提时间对多糖得率影响依次减小,比较分析了各因素条件下松花粉多糖提取得率,为更好利用松花粉多糖提供科学基础。
The condition of extracting water - soluble polysaccharide from massonpine pollen through orthogonal experiment were investigated: ratio of solid- liquid was 1 : 15, temperature was 85℃ , extracted 2 hours, amount to 4 times, decolored through pocket resin, vacuum concentration, got rid of glycoprotein by pepsin-Sevage, deposited it by ethanol whose Liquor precision was 95% and got the impure polysaccharide, measured the absorbency under 490nm, calculated the content of total sugar. Under the conditions, the production rate of polysaccharide amounted to 1.70%, the effect of temperature, ratio of solid-liquid ,times and time on the extraction percent of polysaccharides reduced in turn.
