摘要
以南阳酵母菌为出发菌株,采用紫外线对其原生质体进行物理诱变和TTC法初筛,并以甜高粱秆汁为发酵底物进行发酵试验,最终选育出高产酒精的NY-09菌株。试验结果表明,经紫外线照射20s~30s,致死率为78.7%~86.56%,发酵48h后发酵液残糖为2.09g/mL,酒精度为11.2%,较出发菌株提高了23.3%。经过连续转接10代,NY-09的发酵性能无明显变化,表现稳定。
The protoplast of Nanyang yeast was selected as original strain in this paper. First of all its protoplast was mutated by UV-rays which was a kind of physical mutation, after that TTC method was used as the first selection and at last through the fermenting test of the sweet sorghum stalk juice, we got a strain NY-09 which could exhibit a higher liquor yield rate, Through the result we known that mutated by UV-rays 20 ~30 s, killed rate was 78,7% to 86,56%, after fermented 48 h, The alcohol content reached 11.2% to 23.3% higher than the original stain. After switched over 10 generation, the fermentation by NY-09 didn't show significant changes.
