黄姜皂素生产废渣固态发酵纤维素酶产酶条件优化及酶学特性初探 被引量:5

摘要 以黄姜皂素生产废渣为原料,考察了不同培养条件对木霉产纤维素酶的影响,并进行了酶学性质的初步研究。结果表明,表面活性剂吐温-80添加量为0.08%时能显著提高酶活力;一定量的碳酸钙能显著提高酶活力,其最适添加量为0.5%;通过正交试验找出了最佳的培养条件,CMC酶活最佳培养条件为:培养温度28℃,培养4 d,起始pH5.0。滤纸(FPA)酶活为:培养温度28℃,培养5 d,pH5.0。该酶的最适反应温度为60℃,最适反应pH为4.5,其热稳定性不好,金属离子对其酶活具有抑制作用。 This text used solid waste of diosgenin production as the culture medium, observed and studied the cellulase characteristic. The result showed that when concentration of the surface active agent Tween-80 is 0.08% can enhance the enzyme activity obviously,acertain discharge of CaCOa can also enhance the enzyme activity markedly, the optimal concentration is 0.5 % ; By way of orthogonal test found out the optimal culture conditions, the optimal conditions of CMC enzyme activity: culture temperature 28℃, culture time 4 d, pH5.0. The optimal conditions of FPA enzyme activity: culture temperature 28℃, culture time 5 d, pH5.0. The optimal temperature of reaction is 60℃, the optimal pH of reaction is 4.5, the thermal stability is not well, some metallic ion can restrain enzyme activity.
