摘要
研究一种改良的反相高效液相色谱法测定谷胱甘肽巯基含量。结果表明,采用色谱柱Kromasil-C18(250×4.6mm,5μm),流动相A(0.1%TFA)和流动相B(甲醇)梯度洗脱:流动相B40% ̄80%,0 ̄10min,然后80%B保持5min,流速0.8mL/min,检测波长327nm,得到NTB标准曲线y=3.67059x+0.14123,回收率101.9%,RSD=1.17%,从而建立了一种高灵敏度谷胱甘肽巯基检测方法。
An improved method of determination glutathione sulfhydryl groups by RP-HPLC was investigated. Results showed that the standard curve of NTB was y=3.67059x+0.14123, the average recovery was 101.9%, RSD was 1.17%. Determination parameter was showed as: column, Kromasil-C18, 250×4.6mm, 5μm; flow rate, 0.8mL/min; detection, UV absorbance at 327nm; solvent A,TFA 0.1%(v/v); solvent B, methanol; elution, linear gradient 0~10min 40%~80% solvent B, then, retaining 5min. It was a high sensitivity method to determinate sulfhydryl of glutathione.
