摘要
以1039,B-17和637大白菜自交系为试材,研究了培养基中不同pH和琼脂糖质量浓度对胚状体诱导的效应。结果表明,当pH值为6.0-6.2,琼脂糖质量浓度为8-9 g/mL时,可以诱导“1039”大白菜试材产生胚状体,其诱导频率约1.0%;当pH值为5.7-5.8,琼脂糖质量浓度为8-10 g/mL时,3个大白菜试材中均有不定芽产生,并且随着琼脂糖浓度的增加,不定芽的数量增加。不同的大白菜试材间诱导频率没有明显差异,均为3.0%。
Through changing pH value and agarose concentration of culture medium, we developed a method for embryo and adventitious bud induction. The MS medium supplemented with 6-BA 5 mg/L, NAA 0.5 mg/L, pH 6.0 - 6.2, agrose 8 - 9 g/mL was found to be optimum for embryo induction in 1039 imbred line, and the frequency of induction was about 1.0%. pH 5.7 - 5.8, agrose 8 - 10 g/mL was found to be optimum for adventitious bud inducing in 1039, B- 17 and 637 imbred lines, and with the increase of agarose concentration, frequency of induction also enhanced. The average frequency of adventitious bud induction was about 3 %, indicating no significant difference among our tested lines.
