摘要
背景与目的:检测T细胞非霍奇金淋巴瘤(T-NHL)中ZAP-70的表达水平变化,分析ZAP-70与P53、Ki67的相关性,探讨ZAP-70蛋白在T-NHL中表达的意义。材料与方法:采用SP免疫组织化学方法检测48例T-NHL中ZAP-70、P53、Ki67蛋白的表达,利用χ2、t检验、Spearman's等级相关检验方法分析3种蛋白表达的相关性。结果:ZAP-70在T-NHL中的表达阳性率为58.3%。肿瘤恶性程度越高,ZAP-70蛋白的表达水平越低,ZAP-70蛋白与肿瘤的恶性程度呈负相关关系(P=0.001);P53、Ki67蛋白与肿瘤的恶性程度呈正相关,P53蛋白的表达与Ki67蛋白之间存在相关性(P<0.01),且P53蛋白与EB病毒感染呈正相关(P<0.05)。结论:ZAP-70蛋白表达的缺失对于恶性T细胞淋巴瘤的诊断可能具有一定的辅助作用;联合检测P53与Ki67蛋白的表达水平对判断T-NHL的预后,指导术后辅助治疗具有重要意义。
BACKGROUND & AIM:Assessed the expression of ZAP-70 in T-NHL, and analyzed the correlation between ZAP-70 and P53, Ki67, to explore the significance of ZAP-70 protein expression in T-NHL. MATERIALS AND METHODS: Evaluated the expression of ZAP-70, P53 and Ki67 in 48 T-NHL by SP immunohistochemistry techniques. The results were analyzed by χ^2, t test and Spearman' s rank correlation. RESULTS: Expression of ZAP-70 was detected in 58.3% T-cell lymphomas, and the expression of this protein was correlated negatively with tumor grade. Expression of ZAP-70 in high-grade T-NHL was significantly lower than that in low-grade T-NHL(P = 0. 001).Of 48 T-NHL,expression of P53 was not only correlated positively with tumor grade, but also had a positive relationship with infection of EBV (P〈0.05) .The expression of ki67 was also increased in high-grade T-NHL. Statistical analysis demonstrated that there was a positive correlation between P53 and Ki67 (P 〈 0.01) . CONCLUSION: Absence of ZAP-70 may be diagnostically useful in paraffin sections suggesting T-NHL. Simultaneous measurement of P53 and Ki67 may have great importance in evaluating omgnosis and directing therapy after operation in T-NHL.
