摘要 目的研究新试剂2-[2-'(5-硝基-吡啶)-偶氮]-变色酸(5-NO2-PACA)的合成方法。方法以5-硝基-2-氨基吡啶为原料,经重氮化,再与变色酸偶联合成新试剂,利用试剂与钴在pH=10.00的缓冲介质中的显色反应进行钴(Ⅱ)含量测定。结果得到紫红色晶体,并用元素分析、红外光谱和氢核磁共振谱鉴定了其结构;试剂与钴形成1∶1稳定绿色配合物,λmax=680 nm,ε=8.07×10^4L/mol·cm,钴的测定线性范围为0-10μg/25 mL。结论该法简单易行,准确可靠,可以用于VB12针剂中钴(Ⅱ)含量的测定。 Aim To synthesize 2-[ 2'-(5-nitro-pyridylazo) ] -Chromotropic acid ( 5-NO2-PACA) and develop the newmethod of determination of cobalt( Ⅱ ). Methods Using 5-nitro-2-nitylpyridine as raw material, and by diazoiation, then was coupled with chromotropic acid to synthesize new ingrain of 2- [ 2'- (5-nitro-pyridylazo) ] -chromotropic acid. The method of determination of cobalt was developed based on the reageng with Co ( Ⅱ ) to color reaction in the pH = 10. 00. Results The amaranth complex was produced and the structure of the reageng was char- acterized by elemental analysis and infrared absorption spectrometry. The reageng with Co ( Ⅱ ) to form a stable 1: 1 green complex. The apparent molar absorptivity was 8.07×10^4L/mol·cm at λmax, = 680 nm. The ranges of determination are 0 - 10 μg/25 mL for Co( Ⅱ ). Conclusion The method has been applied satisfactorily to the determination of Co( Ⅱ ) in the injection of VB12.
