摘要
目的:观察阿托伐他汀对大鼠脑缺血再灌注脑组织bcl-2和bax表达的影响。方法:Wistar大鼠随机分为正常组、假手术组、缺血再灌注组(IR组)和阿托伐他汀治疗组(MT组)。采用线栓法复制大鼠脑缺血再灌注模型,应用免疫组织化学法检测再灌注12h和24h大鼠脑组织bcl-2和bax的表达。结果:与正常组和假手术组比较,IR组和MT组bcl-2、bax表达均显著增高(P<0.01);与IR组比较,MT组bax表达显著降低(P<0.05),bcl-2表达增高(P<0.05)。结论:阿托伐他汀能抑制大鼠脑缺血再灌注后脑组织bax和促进bcl-2表达。
Objective To observe the impact of atorvastatin on the expressions of bcl-2 and bax in the cerebral tissues of vascular dementia rats and to study the neuroprotection mechanism of atorvastatin. Methods Wistar rats were randomly divided into normal group (group N), sham-operation group (group S), cerebral ischemia-reperfusion model group (group M), and atorvastatin treatment group (group MT). Expressions of bcl-2 and bax were detected by immunohistochemistry. The value of light density was determined by imaging analysis. Percentage of the cells with expressions of bcl-2 and bax was measured under the light microscope. Results The expression of bax was lower in group MT than in group M (P 〈 0.05) but that of bcl-2 was higher (P 〈 0.05). The expressions of bcl-2 and bax in group MT were higher than those in group N or S (P 〈 0.05) as significantly were those in group M (P 〈 0.01). Conclusion Atorvastatin can decrease bax expression but increase bcl-2 expression in vascular dementia rats, suggesting that it may play a protective role for the cerebrum of ischemic/reperfused rats through inhibition of apoptosis.
