pH吸附法纯化戊糖乳杆菌素31-1的研究 被引量:4

摘要 戊糖乳杆菌素31-1,由分离自宣威火腿的戊糖乳杆菌31-1产生,随pH的改变可以选择性的吸附于产生菌细胞表面,pH5.0时吸附率达75%,而在pH3.0以下,7.0以上吸附率为0。基于以上规律,通过调整pH值,戊糖乳杆菌素31-1在pH5.0吸附、pH7.0释放于产生菌细胞,达到了较好的分离纯化效果,戊糖乳杆菌素31-1回收率为45%,纯化倍数为33。经Tricine-SDS-PAGE进行分子量的估测及纯度鉴定,证实戊糖乳杆菌素31-1的分子量在6400~14000D之间。该纯化方法简便、高效,具有工业生产的潜力。 Pentocin 31-1, produced by Lactobacilluspentosus 31-1 isolated from the traditional China fermented Xuan-Wei Ham, coud be adsorbed selectively on its producer at different pH conditions. At pH5.0, the adsorption rate was 75%, while below pH 3.0 or above pH7.0, Pentocin 31-1 wasn't adsorbed at all. On the basis of this property, by changing pH, Pentocin 31-1 was adsorbed at pH5.0 and released at pH7.0. The efficiency of isolation and purification was high. The specific activity of Pentocin 31-1was increased 33 fold with a yield of 45%. The active extract was further subjected to Tricine-SDS-PAGE. The result showed that two bands appeared, where most of the activity corresponded to the band with molecular weight between 6400- 14000 D. It is simple and effective, could be used as commercial preparations for food industry.
