摘要
目的:建立泽泻中活性成分的高效液相色谱-电喷雾-质谱分析方法。方法:采用Zorbax SB-C18色谱柱;乙腈-水二元梯度洗脱;Agilent电喷雾离子阱多级质谱仪;正离子检出模式。结果:在正离子检出模式下,泽泻的总离子流色谱图特征性很强,通过质谱中的主要碎片对泽泻醇B、泽泻醇B-23-乙酸酯、泽泻醇C-23-乙酸酯、泽泻醇C、16-氧化泽泻醇A、11-去氧泽泻醇C6种主要成分进行了结构解析。结论:泽泻的总离子流色谱图比紫外色谱图具有更强的特征性,在该试验条件下能够同时分析泽泻中的多种活性成分,是一种较好的泽泻药材的质量控制方法。
AIM: To develop a high performance liquid chromatography-electrospray ionization-mass spectrometry (HPLC-EMI-MS) analysis approach of Alisma orientalis (Sam.) Juzep. METHODS: A Zorbax SB-C18 column was used under the gradient elution condition with acetonitrile-H2O. The MS detection was performed on an Agilent ion trap mass spectrometer with an elctrospray ionization source under the positive ion detection mode. RESULTS: Alisol B, alisol B-23-aceate, alisol C-23-aceate, alisol C, 16-oxoalisol A and 11-deoxyalisol C were identified by their MS and MS2 fragments. The MS2 fragments of the 6 compounds were completed. CONCLUSION : The total ion current chromatogram of Alisma orientalis ( Sam. ) Juzep. was more characteristic than of the UV chromatogram. 6 bioactive compounds can be simultaneously analyzed by the HPLC-EMI-MS method established in this experiment and it is a good quality control of Alisma orientalis (Sam.) Juzep.
