摘要
对FTA滤膜用于肉中金黄色葡萄球菌PCR直接检测(无需增菌)模板制备的具体方法进行了研究。结果表明,使用FTA滤膜制备模板时,FTA滤膜吸附样品,干燥后采用10%SDS煮沸该滤膜,可消除结合在FTA滤膜上的PCR抑制因子,采用该方法制备的模板可有效地扩增出目的基因。因此,使用FTA滤膜法制备模板DNA,为快速检测食品中的致病菌构建了一个技术平台。
The methods of extracting DNA (without enrichment) from Staphylococcus aureus in meat with FTA filter were studied. The results showed that when the FTA filter diaphragm preparation template was used, the FTA filter could adsorb sample. Boil the filter using 10% SDS after the sample was dried, which may eliminate the inhibition effect of some factors in FTA filter. This method could be used in the preparation of the template to determined possible effects in increasing the aimed gene. FTA filter using template DNA Preparation for rapid detection of food pathogens could be used to establish a technical platform.
