异丙酚预处理对拟AD模型大鼠学习记忆功能和海马损伤表达的影响 被引量:2

摘要 目的观察异丙酚预处理对拟AD模型大鼠的学习记忆功能和海马损伤神经原纤维缠结(neurofibrillary tangles,NFT)和bcl-2表达的影响,分析异丙酚的脑保护作用。方法大鼠海马CA1区微量注射冈田酸(Okadaic acid,OA),建立拟AD大鼠模型。异丙酚预处理组大鼠于拟AD模型制作前30 min,腹腔注射异丙酚100 mg/kg。然后用Morris水迷宫观察大鼠行为学变化,Bielschowsky染色观察NFT及免疫组化方法观察bcl-2表达的变化。结果与对照组大鼠相比,模型组大鼠学习记忆能力显著降低;Bielschowsky染色观察海马CA1区域出现大量NFT的特征性病理变化,免疫组化bcl-2表达明显减少(P<0.05)。与模型组大鼠相比,异丙酚预处理组大鼠学习记忆能力明显提高;海马CA1区NFT明显减少或消失,免疫组化bcl-2表达明显增多(P<0.05)。结论拟AD模型建立前30 min异丙酚预处理能改善拟AD模型大鼠学习记忆能力,能降低NFT的表达,增高bcl-2表达,有明显的脑保护作用。 Objective To study the effects of propofol pretreatment on learning and memory ability,and to investigate the expression of neurofibrillary tangles( NFT ) and bcl-2 of hippocampus tissues in Alzheimer's disease-like model rats, and to analyze the protective effects of propofol on brain. Methods Okadaic acid ( OA) was injected into hippocampal CA1 region of rats in model group and pretreatment group to establish Alzheimer's disease-like model. Rats in the pretreatment group were abdominally injected with propofol ( 100mg/kg) before making AD-like model. The learning and memory abilities of rats were assessed through Morris water maze behavioral test. NFT were the characteristic pathological changes of AD, examined by Bielschowsky stain of hippocampus, and the expressions of bcl-2 were examined by immunohistochemistry in hippocampus CA1 region of the rats. Results Compared with control group rats, the abilities of learning and memory decreased. Many NFTs were observed in hippocampal CA1 region. The expression of bcl-2 in the CA1 region decreased ( P 〈 0. 05 ). Compared with model rats, the abilities of learning and memory of propofol group rats improved. NFT was reduced in hippocampal CA1 region. The expressions of bcl-2 in the CA1 region increased ( P 〈 0. 05 ). Conclusion Treated with propofol at 30 minutes before injecting OA can ameliorate the neurobehavioral score of rat with hippocampus injury, reduce the level of NFT and increase the expression of bcl-2 in brain tissues of rats with OA injury,which has significant protective effects on brain.
