摘要
[目的]研究在非水相介质中脂肪酶催化丁基苯酞的转酯化可行性。[方法]以脂肪酶Novozyme435、PPL、PS、PS-C和AY为试材,通过酶法反应试验和对反应后丁基苯酞的转化量进行气相色谱分析,研究了某些关键因素如四氢呋喃(THF)、有机溶剂、含水量以及丁基苯酞与乙酸乙烯酯的摩尔比等对转酯化反应的影响。[结果]THF有助于改善水在疏水性介质中的均匀性分布,增加水在疏水性溶剂中的溶解度而降低反应介质的水活度,进而大大提高丁基苯酞在微水相有机溶剂中的转酯化得率。在该反应中,由65%(mol/mol)的THF和35%(mol/mol)的正己烷组成的二元混合溶剂为适宜的反应介质,最适含水量为0.4%(V/V),在该条件下,Novozyme435催化的转酯化可使丁基苯酞的转化率达到49.4%。[结论]脂肪酶Novozyme435对催化丁基苯酞的转酯化具有较高的催化活力。
Objective The aim was to study the feasibility of catalyzing the transesterification of 3-N-butylphthalide by lipase in nonaqueous media.Method With lipases Novozyme 435,PPL,PS,PS-C and AY as tested materials,the influences of some key factors such as THF,organic solvent,water content and molar ratio of 3-N-butylphthalide to vinyl acetate ester on transesterification reaction were studied through enzymatic reaction test and gas chromatography analysis on the transform amount of 3-N-butylphthalide after reaction.Result THF helped to improve the homogeneous distribution of water in hydrophobic media,and increase the solubility of water in hydrophobic solvent,and decrease the water activity of reaction media,and then enormously increased the transesterification yield of 3-N-butylphthalide in microaqueous organic solvents.In the reaction,the binary mixed solvent consisted of 65 %(mol/mol)THF and 35 %(mol/mol)hexane was the suitable reaction media,with the most suitable water content of 0.4 %(V/V).Under the condition,the transesterification catalyzed by Novozyme 435 could make the transform rate of 3-N-butylphthalide reach 49.4 %.Conclusion Lipase Novozyme 435 had higher catalyzing activity on the teresterification of 3-N-butylphthalide.
