不同浓度高碘水对小鼠甲状腺形态和功能影响的实验研究 被引量:19

Effect of different excess iodide on function and morphology of thyroid in mice

摘要 为了解不同浓度的高碘水对小鼠甲状腺功能和形态的影响及其与高碘甲状腺肿发病机理的联系,实验分4个组。对照组(A),小鼠饮水含碘量为10.8μg/L,实验组(B、C和D)小鼠饮水含碘量分别为5000μg/L、10000μg/L、20000μg/L。实验时间为150天。观察甲状腺组织形态学、甲状腺组织碘含量、甲状腺过氧化酶(TPO)活性及血清TSH和甲状腺激素浓度的改变。结果显示实验各组甲状腺组碘含量、血清FT4水平明显高于对照组,且随摄入碘量的增加而增加。实验各级TPO活性、C组和D组血清FT3水平明显低于照组。实验各组甲状腺呈典型的胶质性甲状腺肿。结果表明高碘对小鼠甲状腺形态与功能具有抑制作用。高碘甲状腺肿是由于滤泡扩张,过多胶质堆积在滤胞腔内所致,并非TSH升高导致甲状腺滤泡增生的结果。 The experiment was designed to study pathogenesis of iodide goiter by performin morphological observation and measuring thyroid function.The mice of experimental groups(group B,C and D)were raised high iodine water containing iodine 5000,10000 and 20000 μg per liter respectively.The mice of control group(group A)wer drunk 10.8μg/L water.After 150 days,the mice were killed and studied for morphology of thyroid tissue,thyroid iodine contents,thyroid peroxidase(TPO) activity and serum thyroid hormone and TSH concentrations.The results showed thyroid iodine contents and FT 4 levels of three experimental groups,increased with increasing intake of iodide,were higher than thosse of the contol,but the TPO activity of three experimental groups and FT 4 levels of group C and D was lower than that of the control.The thyroid in three experimental groups was typical colloid goiter.Based on above results,we thought that excess iodine had inhibitory effects on thyroid morphology and function,and iodide goiter was due to excess iodine to form excess thyrocolloid stored in expansive follicular lumens.Iodide goiter was not due to elevation of TSH level to induce the thyroid follicular hyperplasia.Excess iodine could damage the thyroid.
