摘要
目的观察肝细胞生长因子(HGF)和1,25-(OH)2VitD3对兔骨髓基质干细胞(bone marrow stromal cells,BMSCs)成骨活性的诱导作用。方法采用HGF和1,25-(OH)2VitD3联合诱导兔BMSCs后检测其碱性磷酸酶(alkaline phosphatase,ALP)活性、骨钙素和胶原蛋白I、II的含量并观察其成骨分化特性的变化。结果HGF和1,25-(OH)2VitD3联合诱导的兔BMSCs的ALP活性、骨钙素水平和胶原蛋白I、II在细胞中表达水平均明显高于对照组细胞。结论HGF和1,25-(OH)2VitD3联合诱导作用可以促使兔BMSCs明显表现出成骨分化特性。
Objective To investigate the oseogenic capacity of rabbit BMSCs (bome marrow stromal cells) induced by HGF (hepatocyte growth factor) and 1,25 - (OH)2VitD3. Methods the rabbit BMSCs was cultured in vitro and induced by HGF and 1,25 - (OH)2VitD3, then the osteogcnic differentiation capacity had been cultured by detecting the activation of ALP (alkalie phosphatae ) and the expression of osteocalcin and collagen Ⅰ and Ⅱ. Results The activation of ALP and expression of osteocalcin and collagen Ⅰ and Ⅱ of BMSCs induced by HGF and 1,25 - (OH) 2 VitD3 were obviously higher than those of the control at different experiment time ( P 〈 0.01 ). Conclusion the ostengenic differentiation capacity of BMSCs could be induced by HGF and 1,25 - (OH)2VitD3.
