摘要
目的:建立柑橘黄酮中总黄酮及橙皮苷的含量测定方法。方法:采用分光光度法,以芦丁为对照品,测定总黄酮的含量,检测波长为500nm。采用高效液相色谱法测定橙皮苷的含量,色谱柱:Agilent ZORBAX C18(150mm×4.6mm,5μm);乙腈-0.1%磷酸(22:78)为流动相,检测波长为284nm,流速1mL/min。结果:分光光度法测定柑橘总黄酮在8.048~48.288mg/L(r=0.9999)的浓度范围内线性关系良好,加样回收率为101.9%。HPLC法测定橙皮苷在0.01608~0.402g/L(r=1.0000)范围内线性关系良好,加样回收率为101.6%。结论:两种方法均准确、重现性好,适用于柑橘黄酮的质量控制。
Objective :To establish methods for the determination of total flavonoids and hesperidin in Citrus Flavonoids. Methods: UV spectrophotometry was used to determinate total flavonoids. In this method, rutin was used as the reference substance and UV detection wavelength was at 500nm. Hesperidin in Citrus Flavonoids was determinated by HPLC with an Agilent ZORBAX C18 ( 150mm × 4.6mm,5μm) column. The chromatographic conditions for hesperidin included the mobile phase consisting of acetonitrile -0. 1% phosphoric acid water solution (22:78) at the flow rate of 1mL/min, and UV detection wavelength at 284nm. Results :A good linear relationship at the range of 8. 048 - 48.288 mg/L( r = 0. 9999)was obtained while determining total flavonoids with UV spectrumetry and the average recovery was 101.9%. The same result was obtained while determining hesperidin by HPLC. It showed a good linear relationship at the range of 0.01608-0.402g/L( r = 1. 0000) and the average recovery was 101.6%. Conclusion :These methods are accurate, reproducible and can be used to control the quality of Citrus Flavonoids.
