MBL免疫治疗结核病小鼠的实验研究

摘要 目的观察甘露聚糖结合凝集素(mannan-bindinglectin,MBL)对结核分枝汗菌感染小鼠的免疫治疗作用。方法将BALB/c小鼠18只随机分成MBL免疫治疗组、微卡治疗组和结核菌感染对照组.每组6只。小鼠感染结核分枝杆菌.感染2周后分3次分别腹腔内注射MBL、微卡或生理盐水。治疗结束后第3周处死小鼠,观测小鼠体重以及肺、脾脏湿重,观察肺脏病理改变,取肺、脾组织进行结核菌培养和菌落计数。结果治疗后第3周,治疗组小鼠体重和脾脏重量高于对照组,肺脏重量低于对照组,且肺部病变较轻。MBL减轻结核病小鼠体重下降.减少结核病小鼠肺和脾脏结核菌数量。结论MBL对结核病小鼠具有一定的免疫治疗作用。 Objective To investigate the immunotherapeutic effects of MBL(mannan-binding lectin) on murine infection with mycobactefium tuberculosis. Methods Eighteen BALB/c mice were divided randomly into three groups , group 1 treated with mannan-binding lectin , group 2 treated with vaccae, and group 3 treated with PBS ( n =6) . 1 ×10^6 bacili in a volume of 300μL PBS were injected into the tail vein of mice in each group . The MBL.Vaccae(freeze-dfied M, vaccae Vaccine for therapy) and PBS were administered intrapefitoneally divided into three times respectively 2 weeks after infection. The animals were sacrificed in third week after the end of treatment . The weight of mouse , lung and spleen were measured. Then the lung and spleen of mice were proceed to histological examination. The numbers of viable bacteria in lung and spleen were counted. Results The body weights of mice in treatment groups were higher than that in control group respectively ,the lung weight and the number of viable bacteria in lungs and spleens of mice in treatment groups were lower than that in control group ,respectively.MBL could improve host defence against mycobacterium tuberculosis and reduce mycobacteria outgrowth on murine with tuberculosis .Conclusions MBL has the effects of immunotherapy on murine with tuberculosis.
