摘要
目的观察低氧微环境对人外周血T细胞生存活性及其CCR7表达的影响。方法采用密度梯度离心和尼龙毛法分离健康捐献者外周血单个核细胞(PBMC)中的T淋巴细胞,分别置常氧(21%O2)和低氧(1%O2)环境培养16 h。倒置显微镜观察T细胞形态,流式细胞术检测T淋巴细胞凋亡率及其CD4+和CD8+T细胞亚群表面CCR7的表达。结果常氧与低氧环境下T淋巴细胞的形态无明显差异。低氧环境下T细胞凋亡率12.82%±1.36%,明显高于常氧环境下的2.56%±0.98%;P<0.05。低氧与常氧环境下CD4+T细胞表面CCR7平均荧光强度分别为53.50±8.78,71.63±8.50;CD8+T细胞表面CCR7平均荧光强度分别为126.46±11.74,175.84±10.90,P均<0.05。结论低氧微环境可下调人外周血CD4+与CD8+T细胞CCR7的表达,加速T细胞凋亡。
[ Objective] To investigate the effects of hypoxia on CCR7 expression and survival activity of human peripheral blood T lymphocytes. [ Methods] Peripheral blood mononuclear cells (PBMCs) were isolated with density gradient centrifugation, then harvested T lymphocytes by using nylon column. The morphologic characters were by inverted microscope, and detected the survival activity and surface CCR7 expression of CD^4+ and CD^8+ T lymphocytes were detected by flow cytometry on normoxic and hypoxic condition. [ Results] The morpbologic characters of T lymphocytes had no significant difference under normoxic and hypoxic condition. Compared with the normoxic condition (2. 56% ± 0. 98% ), hypoxic condition significantly up-regulated the percentage of apoptosis ( 12. 82% ± 1.36% ; P 〈0. 05). The CCR7 expressions on the CD4^+ and CD8^+ T lymphocytes were both decreased compared the hypoxic condition (53.50 ± 8.78 / 126. 46 ± 11.74) to normoxic condition (71.63 ± 8.50 / 175.84 ± 10. 90; P 〈0.05). [ Conclusion] The hypoxic condition can down-regulate the CCR7 expression of CD4^+ and CD8^+ T lymphocytes, and promote their apoptosis processes.
