摘要
目的研究17β-雌二醇(E_2)对切除双侧卵巢(OVX)大鼠脑缺血区VCAM-1的表达和CD_8^+淋巴细胞活化、浸润的影响。方法利用42只SD大鼠制作大脑中动脉缺血再灌注(MCAO/R)模型,缺血90min后再灌注并用E_2治疗3d或7d时制作冰冻切片,行HE染色、TTC染色、VCAM-1和CD_8抗原的免疫组化染色,显微镜下观察染色结果。结果E_2可以减少OVX大鼠脑梗死体积比,减轻缺血后神经组织损害的程度,使残存细胞数目增加,并能抑制VCAM-1和CD_8分子的表达。结论E_2对缺血脑组织具有一定的保护作用,抑制VCAM-1的表达和CD_8^+淋巴细胞的活化、浸润可能是其机制之一。
Objective To study tile effects of 1713-estradiol on lhe expression of VCAM-1 and the infiltration of CD8^+lymphoeyte in the cerebral isehemla and reperfusion model of adult female rats. Methods The middle cerebral artery of OVX rats was occluded for 90 minutes to result in the focal isehemia. Three or seven days later,all subjects were put to death. TTC staining,HE staining and immunohistoehemieal staining were performed. Results 17β-estradiol could decreased the ratio of infaetion volumes and improved the number of residual cells and inhibited the expression of VCAM-1 and the activation and infiltration of CD^8+ lymphocyte. Conclusion Estrogen showed neuroproteetive effect on brain tissues against cerebral isehemia. This may be the results of inhibiting the expression of VCAM-1 and the activation and infihralion of CD8^+ lymphocyte.
