Investigation on histopathological damages of articular growth plate cartilage, Hver and kidney of rats with fluorosis induced by drinking brick-tea in the high altitude areas

摘要 目的观察砖茶氟在高海拔地区对大鼠骺板软骨、肝脏和肾脏组织的损伤作用。方法利用大鼠在高海拔地区进行亚慢性氟中毒实验,通过关节骺板软骨及肝、肾器官的组织病理学改变来观察高海拔地区砖茶氟的毒性作用。结果砖茶组大鼠胫骨近端骺板软骨较薄,骨小梁稀少;骺板软骨细胞变性、形态改变,在增殖层和过渡层之间出现广泛的软骨细胞坏死,坏死部位细胞显著减少,呈现大面积无细胞区;肝、肾改变较轻,肝损害主要为细胞水肿、嗜酸性变性或坏死,细胞膜损伤破裂、核固缩;肾损害主要表现为水肿变性。氟化钠组各器官改变与砖茶组相似,骺板软骨组织以软骨细胞坏死缺失为主;肝、肾组织主要表现为细胞的浊肿变性。结论高海拔饮茶型氟中毒大鼠骺板软骨组织损伤以软骨发育障碍及细胞坏死为主,肝、肾组织的病变主要为细胞的嗜酸性变和坏死.上述组织的病变在砖茶组和氟化钠组无明显差异。 Objective To investigate the damage of articular growth plate cartilages, livers and kidneys of rats induced by fluoride from brick-tea in the high altitude areas. Methods A subchronic test on rats in the high altitude area was carried out to study the toxicity of brick-tea fluoride while the histopathological changes of articular growth plate cartilages, livers and kidneys were observed. Results In brick-tea groups, the growth plate cartilages in metaphysis of tibia were thin and the amount of the trabecular bone was markedly exiguous. The chondrocytes in the growth plate distorted and degenerated. Extensive necrosis was seen between proliferating layer and transitional layer, and the amount of cells was markedly decreased. The relatively mild damage of livers and kidneys was mainly coagulative necrosis of hepatocytes and hydropic degeneration, respectively. Membranes of livers and kidneys were broken, and pyknosis was seen. The damage of organs in NaF group was similar to that in brick-tea group, and extension chondrocytes necrosis and swollen degeneration of liver and kidney cells were seen. Conclusions Cartilage hypoplasia and chondrocyte necrosis has been identified in growth plate cartilage of rats with fluorosis from drinking brick-tea water in high altitude areas, as well as livers hydropic degeneration and kidneys coagulative necrosis, which is not significantly different between brick-tea group and NaF group.
