氟对骨髓基质细胞增殖和成骨能力的影响 被引量:3

Influence of fluoride on abilities of proliferation and osteogenesis of marrow stromal cells

摘要 目的探讨氟对体外培养的大鼠骨髓基质细胞(MSCs)增殖能力与成骨能力的影响。方法体外培养大鼠MSCs。噻唑蓝(MTT)比色法分析不同染氟水平下MSCs的增殖能力;光、电镜下观察不同染氟水平下MSCs成骨转化能力和相应超微结构变化。结果与对照组比较,低水平染氟(0.02mg/L)96h后促进MSCs增殖(P〈0.05);较高水平染氟(0.05~1.00mg/L)72h即可促进MSCs增殖(P〈0.05或〈0.01);高水平染氟(10.00、20.00mg/L)对MSCs具有抑制增殖和促细胞凋亡作用(P〈0.01)。较高水平染氟(2.00~20.00mg/L)时MSCs成骨转化受抑(P〈0.01),较低水平染氟(0.01~1.00mg/L)时MSCs成骨转化增强(P〈0.01)。结论低水平染氟促进MSCs增殖。成骨转化能力增强;高水平染氟抑制MSCs增殖,促进细胞凋亡,抑制MSCs成骨转化能力。 Objective To study the influence of fluoride on abilities of proliferation and osteogenesis of marrow stromal cells(MSCs). Methods MTr was employed to observe the changes of proliferative ability of MSCs treated by a series of concentrations of fluoride in common medium. While MSCs treated by a series of concentrations of fluoride in osteogenic medium, the morphological changes were obtained by optical and eletron microscope. Results Fluoride enhanced MSCs proliferation at such low concentrations as 0.02~ 1.00 mg/L(P 〈 0.05 or 〈 0.01), but inhibited it at higher levels of 10.00 ~ 20.00 mg/L(P 〈 0.01 ). Fluoride could facilitate MSCs osteogenesis at concentrations of 0.01 ~ 1.00 mg/L(P 〈 0.01 ), but inhibit it at 2.00 ~ 20.00 mg/L(P 〈 0.01 ). Conclusion Fluoride at low concentrations can enhance proliferation and osteogenesis of MSCs, and inhibit them at high concentrations.
