Establishment of animal model of invasive pulmonary aspergillosis in immunosuppressed mice

摘要 目的建立侵袭性肺曲霉病(IPA)动物模型。方法以环磷酰胺为免疫抑制剂,造成小鼠免疫抑制,对腹腔巨噬细胞和脾细胞进行计数;双侧鼻孔吸入烟曲霉菌孢子,不同时间点处死小鼠,进行组织培养及病理分析。结果注射环磷酰胺后,小鼠免疫细胞数量急剧下降,腹腔巨噬细胞数及脾细胞数减少,外周血淋巴细胞百分比降低;对IPA模型组的组织培养可见烟曲霉,病理切片可见肺组织大量烟曲霉菌聚积,组织坏死,形成肺脓肿。结论成功建立了小鼠IPA动物模型。 Objective To establish an experimental model of invasive pulmonary aspergillosis in immunosuppressed mice. Methods Immunosuppressed mice were induced by cyclophosphamide intraperitoneal injections. The quantity of peritoneal macrophages and spleen cells were counted and the aspergillus fumigatus conidia were dropped into the nares of each mouse to cause invasive pulmonary aspergillosis. All mice were killed at diferent time points and the tissues were cultured and histopathologically analyzed. Results The number of peritoneal macrophages and spleen cells were decreased obivously afer the cyclophosphamide injection, the percentage of lymphocytes of peripheral blood was declined sharply. A great amount of aspergillus fumigatus and tissue necrosis were observed histopathologically in the lungs infected with aspergillus fumigatus in immunosuppressed mice and pulmonary abscess was formed. Conclusion An experimental animal model of invasive pulmonary aspergillosis is successfully established.
