摘要
目的培养和鉴定兔骨髓间充质干细胞,基因转染后观察hVEGF165在其中的表达。方法体外培养兔骨髓间充质干细胞,以流式细胞仪检测其表面抗原(CD29、CD44、CD45、HLA-DR)的表达。通过腺病毒载体转染hVEGF165后,采用RT-PCR、Western blot法检测hVEGF165在细胞中的表达。结果通过流式细胞仪检测表明培养的细胞为兔骨髓间充质干细胞,RT-PCR法、Western blot法检测结果均表明hVEGF165基因转染兔骨髓间充质干细胞后能在其中表达。结论成功培养兔体外骨髓间充质干细胞,基因转染后细胞能有效表达hVEGF165。
Objective To cultivate and accredit rabbit mesenchymal stem cells , and to detect the expression of hVEGF165 after gene transfection.Methods Rabbit mesenchymal stem cells was acoredited expression of antigen CD29, CD44, CD45, HLA - DR after cultivated in vitro. The constructed plasmids containing right sequence of hVEGF165 were transfected into rabbit mesenchymal stem cells by adenovirus. The expression of hVEGF165 was tested by RT- PCR and western - blot. Results The expression of antigen of cells indicated the cells were rabbit mesenchymal stem cells. The results of RT - PCR and western- blot showed that the transfected cells could express hVEGFI65. Conclusion The rabbit mesenchymal stem cells can successfully express hVEGF165 after gene transfeetion.
