摘要
采用HPTLC以及HPLC分析测定茶多酚在HSCCC两相溶剂中的分配系数,应用16组Oka两相溶剂体系并采用配方试验设计进行改进.结果表明,利用HSCCC分离茶多酚的较优条件为:环境温度20℃,转速750 r/min,固定相正己烷:乙酸乙酯(体积比)=1:5,流动相速率2 mL/min,初始流动相水,120 min时转换为甲醇:水=1:5进行梯度洗脱.
By using HPTLC to determine allot proportion (KD) of catechins in mutual of solvents, 16 group of Oka menstruums are used in this study and formula experiment design optimized the experiment. The HSCCC suitable condition of catechins separation is that the surroundings temperature is 20℃, rotation speed is 750 r/ min, the fixture is Hexane:Ethyl acetate = 1:5, the preliminary fluxion is water, and switched to Methanol: water = 1:5 at 120 min, the speed of fluxion is 2 mL/min. At the same time, one practical tactic of choosing HSCCC solvent system was provided.
